So I just plugged the hole and according to the spray bottle and pressure gauge, it is holding well. The screw did go all the way through, so it's a good thing I plugged it. However, I notice a second piece of debris stuck in the tire, and pulled it out as well. The tire is not leaking from where I pulled it so it obviously didn't go all the way through. Should I try and plug that spot as well, or leave it since it hasn't been punctured?

Grab some water with a little dish soap in it (or use yucky spit) to see if air bubbles out. If not, then all is well.
That's what I did, and it wasn't leaking. I'll be keeping an eye on it.

I was in the same boat with one of my RS3s! I took it to the MTS (Mexican Tire Shop), and they squirted soapy water on it - nothing. Then they dipped it in the Water Bath - still nothing. So the man grabbed his pliers and yanked the "massive" nail out of the tread, and it was all of an eighth-inch long! No fuss, no muss...
I felt so bad I gave the guy a fiver just for screwing with it for 15 minutes... LOL
I bought a Optima Red top battery equivalent (which they told me was better than the optima) from them once. It died one day after the free-replacement warranty on it expired. I took it there and raised hell about it. Essentially I refused to leave until I got a replacement and began announcing my displeasure as each customer walked in the door.
10 minutes later I had a free optima redtop :)
